Cellular mechanisms of impaired mineralocorticoid and glucocorticoid production by endocrine disruptor DDT

چکیده
Developmental exposure to persistent endocrine disruptors is of a great concern. Affection adrenal hormones production by low-dose dichlorodiphenyltrichloroethane (DDT) has been revealed recently. Objective the research – investigation ultrastructural mechanisms impaired mineralocorticoid and glucocorticoid after prenatal postnatal disruptor DDT. Male Wistar rats exposed low doses DDT during development were studied. Aldosterone corticosterone levels in serum measured enzyme-linked immunosorbent assay, histological examination electron microscopy adrenals performed. Pubertal rats, developmentally DDT, demonstrated lowered aldosterone elevated levels. After puberty showed tendency normalization hormones’ production. Morphological less developed zona glomerulosa pubertal period its relative hyperplasia puberty. Microcirculatory disorders focal cell death observed outer fasciculata. Electron cells found signs suppressed secretory activity no significant reduction mitochondria size adult rats. Total number 1 µm2 cytoplasm percent with swollen matrix diminished compared control. Cells inner fasciculata increased functional activity. Age-dependent changes fine structure similar control, but more pronounced. In this way, cellular mechanism prenatally postnatally are insufficient mitochondrion function reorganization mitochondrial apparatus, which occurs period. Mechanisms secretion attributed enhanced cells.

In Vitro Effects of the Endocrine Disruptor p,p’-DDT on Human Follitropin Receptor
BACKGROUND 1-chloro-4-[2,2,2-trichloro-1-(4-chlorophenyl)ethyl]benzene (p,p'-DDT) is a persistent environmental endocrine disruptor (ED). Several studies have shown an association between p,p'-DDT exposure and reproductive abnormalities. OBJECTIVES To investigate the putative effects of p,p'-DDT on the human follitropin receptor (FSHR) function. METHODS AND RESULTS We used Chinese hamster o...
